In 2011, [[Claire Redfield]] is in attendance at a party at the quarters for non-government organization [[TerraSave]], where she also finds newest member and long-time friend, [[Moira Burton]], older daughter of [[Barry Burton]]. Unbeknownst to Claire, however, she is being watched and soon after, a group of masked militants infiltrate the place, taking the attendants hostage. Claire and Moira are injected by the mysterious attackers and abducted with other TerraSave members.

When Claire and Moira awake, they find themselves in an abandoned prison, in individual cells across from each other. Furthermore, strange bracelets have been attached to their wrists, contraptions that they later discover are used by a watching party to measure the wearer's degree of fear. The cell doors open on their own, allowing Claire and Moira to attempt to make their escape. However, they find themselves in for a true trial as their captor notifies them that they are now participating in a study and experiment on human survival.

As they manage to get out of the prison facility, Both were contacted again by their captor who introduces herself as the Overseer who tells the duo to proceed to Wossek as the duo headed to the radio tower. While Claire fixes the radio signal at the top of the tower, Moira gives a desperate call on S.O.S. Claire later discovers that they ended up in a deserted island. The duo now regroup with fellow TerraSave members Gabriel Chavez and Pedro Fernandez at Wossek only to be contacted again by the Overseer who told them that they were injected with a special virus. After Claire gives Gabe the fuel and the battery for the helicopter, the Overseer manages to activate the siren which she lures them out. However, Pedro becomes mutated into a beast after his bracelet glows in red and to attack both Claire and Moira with his drill. The duo are later saved by Neil Fisher as Claire introduces her boss to Moira as Neil recognizes her that she is Barry's daughter.

6 months later, after receiving audio of his daughter's cry for help, Barry Burton, who is a BSAA Advisor, takes on his father duty role to rescue his daughter, Moira from the mysterious island, along the way through the island he meets and teams up with a little mysterious girl, [[Natalia Korda]] who helps him along the way with her special sense abilities.
